HPLC analysis of phosphoramidites using RP or NP conditions

Phosphoramidites are the essential part in the chemical synthesis of oligonucleotides, short fragments of nucleotides and analogues. During the synthesis, errors can occur which is why the phosphoramidites’ purity needs to be closely monitored.
In this application note, it is shown how to analyse phosphoramidites using two different separation modes: reversed phase (RP) and normal phase (NP) chromatography. Due to the complexity of the analytes different interactions can be used in order to achieve acceptable retenetion and resolution.
