Binary prep. HPLC-system for processing large volumes of ureacontaining protein solutions
Often client specific requirements for multipurpose equipment can not be solved with “off the shelf“ solutions. In this specific case, the Kronlab custom build approach proved very useful: every component has been specifically approved for the application at hand and following a specified procedure it is integrated into an LC system. The result is a made-to-measure system.
In this example, which has been designed for a small to medium scale production process, the challenge was to introduce large volumes of sample, which contained
proteins dissolved in urea, onto a reversed phase system. The answer was to implement a “metal-free” HPLC pump, through which the sample could be pumped directly onto the column, using a 4-way 2- position valve. Obviously the size and quality of the pump was chosen in such a way that there was no need to compromise on the speed at which the sample is introduced to the system, compared to the normal flow rate of the system. As this pump can be completely integrated into the system software, the process can be fully automated.
Additionally the use of a diode array UV detector enables simultaneous data acquisition at multiple wavelengths. The inclusion of a software controlled 8 port fraction collector makes it possible to collect fractions based on peak slope, threshold and time to ensure only genuine sample peaks are selected.
Finally Kronlab supplies the customer with detailed documentation of testing as well as qualification or support for IQ/OQ/PQ verification. Individual, client-specific testing and acceptance criteria can be incorporated on request.
