Breaking Barriers in siRNA Delivery

To accelerate the understanding of disease and the development of gene knockdown therapies, Integrated DNA Technologies (IDT) is facilitating the advancement of a new transfection technique.

Breaking-Barriers-siRNA-DeliveryThis nanoparticle-based method is explored in a recent DECODED article contributed by Precision NanoSystems Inc. (PNI). By combining IDT’s highly potent Dicer-substrate RNAs (DsiRNAs) and PNI’s unique nanoparticle delivery system for hard-to-transfect cells, the IDT-PNI partnership presents a successful solution for challenging gene silencing applications, achieving >95% transfection efficiency.

The novel NanoAssemblr™ system and SUB9KITS™ reagent kits from PNI enable the simple assembly of novel nanoparticles using advanced microfluidics, which minimizes waste material and ensures a consistently tuned particle size. These particles are designed to deliver DsiRNA into the cell via a natural pathway, avoiding carrier effects and limitations, such as toxicity and poor performance in postmitotic cells. And the particles greatly enhance the efficiency of RNA transfection, even in notoriously difficult-to-transfect neurons.

Scientists can now focus on studying the effects of gene knockdown, enabling many new and exciting research projects that were previously limited by the low transfection efficiency and/or high toxicity of traditional delivery methods.
