Dionex Demonstrates Fast Desalting of Oligonucleotide and Protein Samples
Dionex is proud to announce a new method to desalt oligonucleotide or protein samples for either on-line analysis by high-performance liquid chromatography (HPLC) or liquid chromatography-mass spectrometry (LC/MS). Application Update 174: On-Line High-Throughput Desalting to Prepare Samples for Mass Spectrometry demonstrates a simple, fast, and effective process that was completed within 2 min, including sample desalting and flushing, for an oligonucleotide sample and a bovine serum albumin (BSA) sample using a Dionex UltiMate® 3000 HPLC system and Chromeleon® Chromatography Data System (CDS) software.
MS has become a powerful tool for the analysis of complex biochemical samples. These samples often contain high concentrations of salts that interfere with the ionization process and may result in lower peak intensities, compromised peptide/protein detection, and frequent contamination of the ion source. Therefore, it is essential to remove salts from samples before subjecting them to MS detection. Although off-line solid-phase extraction (SPE) is the most common desalting method, it is time-consuming. This application update describes a simple, fast, and effective on-line method using an Acclaim® PolarAdvantage II Guard column for high-throughput sample desalting before MS detection.
