ASIR Purification system from Dynamic Extractions Standard Impurity isolation
In medicinal chemistry a challenging aspect to
preparing compounds for testing, as lead molecules, can be their isolation from
the crude reaction mixture that may contain catalysts, salts, metals, and other
components which interfere with traditional purification methods. One
such problematic compound is triphenylphosphine oxide (TPPO). TPPO is notoriously difficult and time
consuming to remove using standard solid phase chromatography methods. In this
example a standard HPCCC method has been developed that targets TPPO,
regardless of the compound of interest's polarity. This enables chemists to rapidly
remove TPPO from crude reaction mixtures and isolate suitable quantities of
high purity target compound(s).
The chromatogram shows the method applied to a reaction workup of a Staudinger reduction of a protected glycosylazide. In this particular example the target compound elutes after TPPO, as it is non-polar relative to TPPO. The TPPO elutes as a single symmetrical, narrow peak; no streaking or tailing occurs. The method is designed so that TPPO will always elute at 31 minutes. The desired target reaction products will elute earlier if more polar or later if more non-polar than TPPO. Total recovery of applied reaction mixture is achievable using this system.
Dynamic Extractions now provides their SII purification system to solve these challenges. Incorporating our Spectrum HPCCC instrument, that is preprogrammed with a number of standard methods for impurity removal. Sample loading from a few milligrams to a gram is possible on this system. The scientist simply loads their sample, selects the impurity to be removed and the system will then deliver the purified material at the fraction collector. The system also incorporates pumps, detection, auto-sampler, fraction collector, valving and software.
