release of Application Update 178:
Faster Solution with Increased Resolution for
Determining Chromatographic Identity and Absence of OSCS in Heparin Sodium. The
U.S. Pharmacopeia Monograph for Heparin Sodium was revised after oversulfated chondroitin
sulfate (OSCS)-contaminated heparin led to the deaths of over 100 patients.
The method demonstrated in AU 178 achieves better resolution of
heparin, dermatan sulfate, and OSCS using the higher-capacity
Dionex IonPac® AS11-HC column, instead of the L61 column (e.g., IonPac
AS11 column) specified in the monograph. In addition to improved resolution,
the method is significantly faster, and it replaces the sodium perchlorate
eluent with a more environmentallyfriendly sodium chloride eluent.
